Leon S. Kennedy is a special agent for the U.S. government and one of the few survivors of the Raccoon City disaster. Ada Wong is an enigmatic spy and mercenary operating for unknown organizations, specializing in infiltration, intelligence gathering, and biological weapon recovery. Her past remains shrouded in mystery, and very few know her true motivations. Following the events in Raccoon City, she continues to get involved in operations related to bioterrorism, always acting in her own best interest. During the mission in Spain, she closely monitors the activities of Los Iluminados and Las Plagas, moving through the shadows.

Ashley Graham is the daughter of the President of the United States. Kidnapped by the Los Iluminados cult, she is taken to a remote rural region of Spain as part of Osmund Saddler's plan to spread Las Plagas and infiltrate the highest spheres of the government. During her captivity, she is infected with the parasite, though she has not yet lost her free will. Her rescue is left to Leon S. Kennedy, with whom she develops a strong bond based on trust and cooperation while they try to survive the horrors surrounding them.

Luis Serra Navarro is a former researcher with a past tied to the development of biological weapons. Born in a small Spanish village, he left his home after his grandfather died, and years later worked as a researcher for the Umbrella Corporation. After discovering the consequences of his research, he turned away from the organization. His knowledge of Las Plagas and the plans of Los Iluminados makes him a key asset during the events of Resident Evil 4, where he decides to help Leon S. Kennedy in an attempt to make amends for his past mistakes.

Ingrid Hunnigan is an intelligence analyst and government field liaison for the United States. Her role is to coordinate high-risk missions from operations headquarters, providing tactical support, up-to-date information, and constant communication with agents deployed in the field. During the Ashley Graham rescue mission in Spain, she acts as Leon S. Kennedy's main contact, assisting in data gathering and helping him adapt to an ever-evolving situation.
